CAPILLARY ELECTROPHORESIS SIZING PRECISION OF MULTIPLEX SHORT TANDEM REPEAT SYSTEMS USING DIFFERENT INTERNAL SIZE STANDARDS AND MATRIX TYPES

Streszczenie
One of the critical elements strongly influencing correct DNA typing using capillary electrophoresis is precision of analysis. Modern commercially available multiplex kits use highly discriminating STR systems containing alleles differing in size by 1 or 2 nucleotides. Confident distinguishing of such alleles requires high precision of analysis. In the case of commercially available multiplex kits, unsatisfactory precision was observed. The main aim of this work was to compare precision and accuracy of DNA typing using commercially available multiplex kits containing complex STR loci with three different DNA internal standards: the GS500, ILS400 and ILS600. The influence of polymer type on accuracy and precision of analysis was also investigated. It was found that application of commercially available internal lane standard ILS600 significantly improves analysis precision.
