SWGDAM VALIDATION STUDY OF THE PROFILER PLUS KIT FOR FORENSIC CASEWORK ANALYSIS

Studies were performed as recommended by the Scientific Working Group on DNA Analysis Methods (SWGDAM previously known as TWGDAM) committee to validate the AmpFlSTR Profiler Plus PCR Amplification Kit for forensic casework applications. Amplification products were detected using the ABIPRISM 310 Genetic Analyser. Various biological traces, stored for various time periods and in different environmental conditions, were examined. They were blood, saliva and semen stains incubated at high temperature and humidity and blood stains on various substrata. The influence of two methods of DNA extraction from biological traces and of various inhibitors on the specificity and efficiency of PCR amplification was also tested. It was shown that the most useful method of DNA extraction from biological traces is the classical phenol-chloroform method, which offers high recovery of pure DNA. It was also observed that the DNA isolation method influences not only the efficiency of DNA extraction, but also the proportion of heterozygous alleles and the reciprocal balance of individual loci of the examined system.
