POLYMORPHISM OF STR SYSTEMS (D9S304, D1S533, D7S820, FGA) IN POPULATION SAMPLES OF SOUTH-EAST POLAND

Four STR systems: D9S304, D1S533, D7S820, FGA were analysed on 301 DNA samples. Amplification was carried out using MultiPlex II kits (Lifecodes). PCR products were separated using polyacrylamide gel electrophoresis and the silver staining method. In the D9S304 locus, 10 alleles were identified (the most frequent of which was allele 8 – 0.377), in the D1S533 locus 10 alleles (the most commonly occurring one was allele 13 – 0.307), 8 alleles in the D7S820 locus (the most frequent was allele 10 – 0.319), and 8 alleles in the FGA locus (with the most frequent allele 22 – 0.219) were observed. The calculated heterozygosity of analysed STR systems was 0.791, 0.804, 0.791 and 0.860 respectively. The distribution of genotype frequencies shows no deviation from the Hardy-Weinberg equilibrium. The high usefulness of the MultiPlex II kit in putative paternity testing was confirmed by the value of the Power of Discrimination coefficient (PD = 99.99), Paternity Index (PI = 51) and analysis of genotyping results in 87 paternity cases.
